ABSTRACT

Objective To explore the mechanism of Xiaoying decoction on experimental autoimmune thyroiditis (EAT) rats in view of regulatory T cells and Th17 cells. Methods SD rats were divided into five groups, normal control group, model control group, tripterygium glycoside group,Xiaoying decoction low and high dose groups.Except for normal control group, the other groups were established the model of EAT.Rats in the Xiaoying decoction low,and high dose groups were given Xiaoying decoction of 17.24 and 68.95 g·kg-1;rats in the tripterygium glycoside group were given tripterygium glycoside 6.25 mg·kg-1.The serum free triiodothyronine(FT3),free thyrocyte(FT4) and thyroglobulin antibody were detected by RIA method.Thyrocyte morphology was observed under optical microscope. The expression levels of Foxp3 mRNA and IL-17 mRNA were detected by real-time PCR.The changes of Treg cells and Th17 cells were analyzed by flow cytometry. Results Compared with the normal control group,FT3,FT4 and TgAb were increased in the model control group (P <0.01,P <0.01,P <0.05).Compared with the model control group,FT3,FT4 and TgAb were decreased in tripterygium glycoside group and Xiaoying decoction high dose group (P < 0.05).The infiltration score in the normal control group, model control group, tripterygium glycoside group,Xiaoying decoction low and high dose groups were 0,4,4,3.5,2.Compared with model control group, the infiltration was improved (P <0.01).Foxp3 mRNA expression was decreased while IL-17 mRNA increased in the model control group as compared with the normal control group(P < 0.01).In contrast, the expression of Foxp3 mRNA was increased and IL-17 mRNA expression was decreased in Xiaoying decoction low and high dose groups, tripterygium glycoside group as compared with the model control group (P <0.05).Compared with the normal control group, rats in the model control group had fewer Treg cells and more Th17 cells (P <0.01).Compared with the model control group, the percentage of Treg cells was elevated and Th17 cells was reduced in tripterygium glycoside group and Xiaoying decoction high dose groups (P < 0.01).Conclusion The therapeutic mechanism of Xiaoying decoction on EAT rats may be related to changing the percentage of regulatory T cells and Th17 cells with up-regulating the expression of Foxp3 mRNA and down-regulating the expression of IL17mRNA.

ABSTRACT

Objective To investigate the clinical efficacy and its mechanism of Yangyin Xiaoying decoction combined with western medicine in the treatment of Graves' Disease. Methods A total of 50 patients suffered from Graves' Disease in our hospital were selected from January 2012 to December 2014,and were randomly divided into observation group and control group using random number table method,25 cases in each group.Other 25 cases of healthy subjects were selected in the same period in our hospital as the healthy control group. The control group received the treatment of methimazole, propranolol and other western medicine. The observation group received Yangyin Xiaoying decoction combined with western medicine. After 2 months of treatment, the levels of free triiodothyronine (FT3),free thyroxine (FT4),thyroid stimulating hormone (TSH),thyroglobulin antibody (TGAb) and thyroid peroxidase (TPOAb),and the percentages of Th1,Th2,Th17 cells in CD4+ cells and their corresponding cytokine levels were detected. Results After treatment, the levels of FT3,FT4,TGAb and TPOAb in the observation group and control group were significantly decreased, and levels of TSH were significantly increased in the two groups (P<0.05);percentages of Th1,Th2 and Th17 cells in the observation group and control group were significantly lower than those before treatment (P<0.05).After treatment, the interferon-γ (IFN-γ),interleukin (IL)-4 levels and IFN-γ/IL-4 ratio,IL-12,IL-5 levels and IL-12/IL-5 ratio in the observation group and control group were significantly lower than those before treatment (P<0.05).The IL-17A,IL-17F and IL-23 levels in the observation group and control group after treatment were significantly decreased, compared with those before treatment (P<0.05).The degree of improvement of above indexes in the observation group were better than those in the control group (P<0.05).Conclusion Yangyin Xiaoying decoction combined with western medicine can significantly improve patients' thyroid function, which suggests that it may have a role in the therapy in the treatment of Graves' disease.

ABSTRACT

Objective To evaluate the therapeutic effect of Xiaoying Ruanjian Decoction on experimental autoimmune thyroiditis (EAT) from the levels of autoantibodies, interleukin-1, tumor necrosis factor-? and apoptosis-regulated proteins Fas/Fasl of EAT rats, and investigate the mechanism from the relation between cytokines and apoptosis-regulated proteins. Methods Sixty SD female rats of SPF level were divided into normal group and model group. An animal model of EAT in rats was developed by injecting thyroglobulin mixed with Freund’s adjuvant immunization and drinking periodate water. The rats modeled were divided into Chinese medicine group, western medicine group and model group. The levels of autoantibodies, interleukin-1, tumor necrosis factor-? were detected with enzyme-linked immunosorbent assay. The level change of Fas/Fasl in the thyroid of rat was observed with immunohistochemical staining. Results Ruanjian Xiaoying Decoction significantly reduced the levels of autoantibody, interleukin-1, tumor necrosis factor-? and lowered the expression of Fas and Fasl in thyroid cells. Conclusion Ruanjian Xiaoying Decoction alleviates the levels of apoptosis-regulated proteins Fas/Fasl by decreasing the relative expression of antibodies and cytokines in serum levels, which plays a role in reducing apoptosis of thyroid cells.
